Abstract

This study aims to analyze and compare the effectiveness of 70% and 96% ethanol solvents in extracting saponin compounds from red seaweed (Eucheuma spinosum) from Panjang Beach, Bengkulu City. Saponin is a secondary metabolite that has important biological activities such as antioxidants, antimicrobials, and anticancer potential for the health sector. Extraction was carried out using the maceration method followed by quantitative analysis of UV-Vis spectrophotometry at a wavelength of 495 nm. The results showed that the crude drug had a drying loss value of 89.36%. The 70% ethanol solvent produced a higher extract yield (22.95%) compared to 96% ethanol (12.45%). However, the determination of saponin content showed different results; the 70% ethanol extract had a higher average content of 89.10% (891 mg DE/g) compared to 96% ethanol which was only 88.51% (885.1 mg DE/g). Statistical analysis of the independent sample t-test showed a significance value of 0.861 and 0.862 (p > 0.05), indicating no significant difference between the two solvents. It was concluded that 70% ethanol is a more optimal and selective solvent in extracting saponin compounds from E. spinosum for the purpose of standardizing natural active ingredients.
