Abstract

Chronic suppurative otitis media (CSOM) is a persistent inflammatory condition of the middle ear characterized by long-standing tympanic membrane perforation and continuous or recurrent otorrhea. The condition is most commonly associated with bacterial infection, particularly Pseudomonas aeruginosa, which plays a major role in chronic disease progression and tissue damage. This experimental study aimed to evaluate the histopathological features of middle ear tissue in a rat model of CSOM induced by P. aeruginosa. The research employed a post-test only control group design using male Wistar (Rattus norvegicus) rats divided into two groups: a Sham group (S1) and an induced group (S2). CSOM was induced through bilateral tympanic membrane perforation followed by transcanal inoculation of 0.1.CSOM induction was performed through bilateral tympanic membrane perforation followed by transcanal administration of 0.1 ,The bacterial inoculation was repeated after one week, and the animals were observed for three weeks. Histopathological evaluation was performed using hematoxylin-eosin staining to assess tympanic membrane thickness, mucosal epithelial damage, inflammatory response, and connective tissue sclerosis. These findings confirm the successful establishment of a reproducible CSOM animal model that can be utilized for further studies on disease mechanisms and therapeutic approaches.
