Abstract

Cardiovascular diseases (CVDs) remain a leading cause of morbidity and mortality worldwide, with increasing incidence among young adults due to unhealthy lifestyles and metabolic risk factors. Current pharmacological therapies face limitations, including suboptimal efficacy and adverse effects, highlighting the need for novel therapeutic agents. Mangiferin, a natural polyphenolic compound derived from Mangifera indica, has recently gained attention for its potential cardioprotective properties. This study aimed to explore the cardioprotective effects of mangiferin and its underlying mechanisms in cardiovascular diseases. This research employed a scoping review approach by systematically identifying and analyzing experimental studies published between 2020 and 2025 from databases such as Google Scholar and ResearchGate. Articles were selected based on predefined inclusion and exclusion criteria, focusing on studies investigating the cardioprotective effects of mangiferin. The findings indicate that mangiferin exerts significant cardioprotective effects through antioxidant and anti-inflammatory mechanisms. It enhances cellular antioxidant defense by activating the PI3K/AKT/Nrf2 signaling pathway, suppresses inflammatory responses by inhibiting NF-kB and cGAS-STING pathways, and reduces apoptosis. Additionally, mangiferin improves endothelial cell viability, decreases oxidative stress biomarkers, lowers pro-inflammatory cytokines, and enhances cardiac function in experimental models. In conclusion, mangiferin demonstrates strong potential as a novel plant-derived therapeutic agent for cardiovascular diseases. Its multi-targeted mechanisms support its role in protecting cardiovascular tissues. However, further clinical studies are required to validate its efficacy, safety, optimal dosage, and bioavailability in humans, as well as to support its development into standardized therapeutic formulations.
